  Synopsis: This article seeks to review the current knowledge about the cross-talk between adenosine A2A receptors (A2ARs) and brain-derived neurotrophic factor (BDNF). A2ARs exert a great influence on the activation of BDNF receptors and on the effect that the neurotrophin has on synaptic transmission. These and other A2ARs functions will be reviewed and discussed in the light of possible A2ARs-based therapeutic approaches in Huntington’s disease, in which a pathogenetic role of BDNF has been shown.

  Synopsis: The signal transduction deficits in Alzheimer’s disease (AD) converge to the receptor-G protein interface, where G protein-coupled receptor kinases (GRKs) act. Beta-Amyloid can decrease membrane and increase cytosolic GRK2/5. GRK2 accumulates near damaged mitochondria and neurofibrillary tangles, and correlates with cognitive decline. The GRK5 deficiency impairs presynaptic M2 desensitization, reduces acetylcholine release, and leads to cholinergic degeneration and cognitive impairment. It also promotes Beta-amyloid accumulation and exaggerates brain inflammation; therefore, it appears to be closely involved in the AD pathogenesis.

  Synopsis: Understanding the underlying molecular mechanisms of synapse formation and development that are associated with learning and memory is of great interest. This article summarizes our current understanding of Kalirin-7 (Kal7) functions, briefly describing how Kal7 regulates spine/synapse formation in hippocampal neurons, and addressing the roles of Kal7 in estrogen- and chronic cocaine exposure–mediated spine plasticity. Kal7 is a key component of mature excitatory synapses and is essential for specific aspects of excitatory synapse formation and function in hippocampal neurons.

  Synopsis: Perturbations in ion gradients that exist across the plasma membrane of living cells have profound effects on cell viability, metabolism, and functional responses. Potassium permeability will influence the proinflammatory responses of macrophages, although the mechanisms remain poorly defined. This report details how inhibitors of a specific potassium ion channel can regulate the activity of macrophage cdk2, a key kinase involved in augmenting the expression of interleukin-6 that is instrumental in regulating an inflammatory response.

  Synopsis: Execution of a perfectly balanced acute inflammatory/reparative response is nowhere as critical and conserved as in the delicate visual axis, as uncontrolled inflammation impairs vision and leads to blindness.   Synopsis: Obesity involves a state of chronic “low-grade” inflammation in adipose tissue, which predisposes to obesity-related comorbidities, such as insulin resistance and nonalcoholic fatty liver disease. Although omega-6 PUFA–derived proinflammatory eicosanoids initially trigger the inflammatory response, in time, there is a switch to the formation of anti-inflammatory and proresolving omega-3 PUFA–derived lipid autacoids, such as resolvins and protectins, which participate in the resolution of adipose tissue inflammation and in preventing the development of obesity-related complications.
